Take the tubes from the ice and place them in a waterbath at 42°C for 50 seconds. Then place them immediately back on ice. This procedure is called heat shock it causes the cell membrane of the bacteria to become permeable and the plasmid is allowed to enter the bacteria. The membrane closes again when the bacteria are placed back in the ice. | ||
